SELECTING THE BEST GEL FOR YOUR SAMPLE

Single Concentration:

Gradient Concentration:

Use low acrylamide concentration gels for larger molecular weight particles.  Increasing the acrylamide concentration decreases the gel pore size.  Single concentration gels are provided with a 4% stacking gel layered on top of the resolving gel, except for gels with catalog designation C1G or C2G, TBE Gels, and TBE/Urea Gels.  A 3% stacking gel is available upon request.  Jule, Inc. will pour any single concentration gel.  Specify the gel you want (click here)!

Gradient gels provide sharper bands and they separate a broader range of molecular weight sizes on the same gel.  Gradient gels have a lower concentration of acrylamide at the top of the gel and the concentration increases uniformly (linearly) to the bottom of the gel.  That is, the gel pore size decreases from the top of the gel to the bottom.  Use the gel photographs below to aid in your selection.  Prices start at $9.95 each for mini gels.  Jule, Inc. will pour any gradient range, however, certain gradient gels have a special low price. The sample run on each gel  above had 13 protein bands, which ranged in size (Daltons) as follows: 212,000 (top of gel),  158,194,  116,351,  97,184,  66,409,  55,561,  42,710,  36,487,  26,625,  20,040,  14,313,  6,517, and   2,340 - 3,400 (Insulin A, B chain) Daltons. Two bands at 66,409 and 26,625 Daltons are double intensity in order to serve as reference points.   Notice that in a low acrylamide concentration gel the smaller protein bands can run off of the gel, but larger proteins sizes are well separated.  In high acrylamide concentration gels the smaller proteins are better resolved.  A gradient gel can resolve large sizes as well as small sizes on the same gel.

The gels above are acrylamide denaturing gels, 0.375 M Tris-HCl, pH 8.8, 0.1% SDS (available with or without SDS), used with the Tris-Glycine running buffer (except for Tricine gels).
